The Human Apolipoprotein A1 (Hu ApoA1) ELISA quantitates Hu ApoA1 in human serum, plasma, or cell culture medium. The assay will exclusively recognize both natural and recombinant Hu ApoA1. Principle of the method The Human ApoA1 solid-phase sandwich ELISA (enzyme-linked immunosorbent assay) is designed to measure the amount of the target bound between a matched antibody pair. A target-specific antibody has been pre-coated in the wells of the supplied microplate. Samples, standards, or controls are then added into these wells and bind to the immobilized (capture) antibody. The sandwich is formed by the addition of the second (detector) antibody, a substrate solution is added that reacts with the enzyme-antibody-target complex to produce measurable signal. The intensity of this signal is directly proportional to the concentration of target present in the original specimen. Apo A1 (Apolipoprotein A1) wird in der Leber und im Dünndarm synthetisiert und durch die Nahrungsaufnahme von Lipiden reguliert. Es ist der Hauptbestandteil von humanen Lipoproteinen mit hoher Dichte (HDL) und spielt eine wichtige Rolle bei der Stoffwechsel- und Transportfunktion von Lipoproteinen im Serum. Es wird durch die Nahrungsaufnahme von Lipiden reguliert und dient als Aktivator für das Enzym Lecithin Cholesterinacyltransferase, das Cholesterin in Cholesterylester umwandelt. Bei der Lipolyse im peripheren Gewebe kann Apo A1 auch eine Rolle als Akzeptor für Oberflächenkomponenten von Lipoproteinen mit sehr geringer Dichte und Chylomicronen spielen. Das Gen, welches Apo A1 kodiert, befindet sich auf Chromosom 11.
